摘要
用正交试验的方法筛选了苏云金杆菌高产苏云金素的培养基配方和培养条件。在第一次正交试验筛选出的6种培养基配方的基础上,对培养基成分和用量作调整后进行第二次正交试验,筛选出五种培养基配方。将这11种培养基用3个菌株继续评选,尽管不同菌株在同一培养基中产苏云金素的量不同,但3个菌株产苏云金素量最高,且稳定的培养基匀为M_3培养基。正交试验的最后培养条件:温度32℃、pH6.5、通气量最大,转速200rpm、接种菌龄16小时、接种量3%、培养24小时。同时,还对苏云金菌杆CT—43菌株在培养基M_3中不同发酵时间的细胞数、pH、还原糖、氨态氮和可溶性磷以及苏云金素的杀虫活性进行了检测。
By the orthogenal test, the medium formulae and incubate conditions for high-yield thuringiensin of Bacillus thuringiensis were selected. Based on the six formulae from the first orthogenal test, the composition of media was adjusted. Then the second test was conducted, and five formulae were obtained from it, The above 11 medium formula were continually selected with three strains of Bacillus thuringienssi, M3 was the best and stablest medium for the three strains to thuringiensin yield.The best incubate conditions are: temperature 32℃, pH6.5, speed of agitation 200rpms 16 hours of seed cultivation; 3 % seed quantity and 24 hour cultivation.When Bt CT-43 strain on the medium M3 were cultured for different time, the abilities of thuringiensin, pHs of medium; the amounts of cell reductive sugar ammoniacal nitrogen and soluble phosphorus were detected.
