摘要
本文评价了夹心 ABC-ELISA 对包虫病人体内循环抗原(HcAg)的检测方法。实验证实,用半饱和硫酸铵盐析的羊源细粒棘球蚴囊液(EgCF)免疫兔产生的抗血清测 HcAg 基木无非特异性反应。用生物素标记抗血清时,其抗体滴度不降,而使夹心 ABC-ELISA 比常规夹心 ELISA 更为优越。在对50例囊型包虫病人体内 HcAg 检测时发现,用酸解血清中循环免疫复合物比末解血清的 HcAg 检出率高一倍(分别为32%、16%)。酸解组及未解组分别检测的 HcAg 为结合型及游离型,二组综合检出率达40%。在对25例猪囊虫病人检测 HcAg 时全为阴性,说明两种蠕虫病间在测抗体时的严重交叉反应在测循环抗原时可完全排除。
The methods of detecting hydatid circulating antigen (HcAg)in human cystic hydatidosis with sandwich ABC-ELISA(Avidin-Biotin-HRP-Complex ELISA) have been evaluated.The hyperimmune rabbit antiserum was prepared by immunization wi- th sheep origin Echinococcus granulosus cyst fluid (EgCF) purified by 50% saturation of ammonium sulphate salting out,and a non-specific reaction could be not found when using the antiserum to detect the HeAd.The titre of the antibody was not decreased when the purified antiserum was labelled with biotin (BNHS).It was found that the positive rate of HcAg in acid-dissociated hydatid circulating immune complexes sera group was higher one time than the one of non-acid-dissociated sera group (32%;16%) in 5o cases of hydatidosis.The acid- dissociated sera group and non-acid-dissociated sera group detected the HcAg of conjugated type and drifted type respectively,and the composite posi- tive rate of two groups was 40%.The 25 cases of human cysticercosis (Cysticercus cellulosae)were negative in HcAd.It is showed that the cross reac- tions of two nelminthiases are very strong when detlecting their antibodies,but the cross reactions are completely eliminated when detecting their cir- culating antigens.
