摘要
采用脂质体介导的基因转移技术,将外源野生型P53基因转染染尘肺泡巨噬细胞(AM)作用下的体外培养的人胚肺二倍体成纤维细胞(2BS),观察其细胞增殖活力及羟脯氨酸含量变化。结果显示:转染P53基因后,2BS细胞的增殖能力明显低于未转染P53的对照细(P<005),羟脯氨酸含量变化不明显。因此提示:野生型P53基因对染尘AM作用下的2BS细胞增殖有一定抑制作用。
Foreign wild type P 53 gene was transfered into human lung fibroblasts in vitro(2BS cells) under the effect of alveolar macrophage(AM) exposed to dust with liposome mediated transfection(lipofection),The proliferation activity and the hydroxyproline(HOP) content of 2BS cells were studied.The results were as follows.After transfection P 53 gene,The proliferation activity of 2BS cells was significantly lowered than that of control group (P<0 05),the HOP content showed insignificant change.It is suggested that foreign wild type P 53 gene might inhibit the proliferation of 2BS cells under the effect of AM exposed to dust,to some extent.
