摘要
探讨了阿霉素中毒心肌细胞脂质过氧化损伤与心肌细胞内钙离子超负荷的关系,并观察了中药生脉散注射液对阿霉素中毒心肌细胞的保护作用。结果发现中毒心肌细胞较正常心肌细胞中超氧化物歧化酶(SOD)活性明显降低,而丙二醛(MDA)、细胞内游离钙浓度明显增高(P<0.01);给予生脉散注射液(3g/L)后,细胞内 SOD 活性明显升高。MDA 及细胞内钙离子浓度明显下降(P<0.05)。结果表明,阿霉素中毒心肌细胞损伤与自由基触发的脂质过氧化反应及其诱导的细胞内游离钙超负荷密切相关;生脉散通过抗脂质过氧化作用逆转中毒心肌细胞钙离子过负荷,可防治阿霉素中毒性心肌损伤。
The present study explored the relationships between the lipid peroxidation injury of the adriamycin-induced toxic myocar- dial cell and the intracellular myocardial Ca<sup>2+</sup>overloading,and observed the protection of the Chinese medicine Shengmai injec- tion(SMI)on the adriamycin induced toxic myocardial cells.The findings suggested that the activity of superoxide dismutase (SOD)was decreased significantly while malonyldialdehyde(MDA)and the intracellular free Ca<sup>2+</sup>concentrations were increased significantly(P<0.01)in the toxic myocardial cells compared with those in the normal myocardial cells.After given SMI(3.0 mg/ml),the activity of the intracellular SOD was increased significantly while MDA and the intracellular Ca<sup>2+</sup>concentrations were decreased significandy(P<0.05).The results were shown that the injury of the adriamycin-induced toxic myocardial cells was closely related to the lipid peroxidation reaction triggered by lipid peroxidation reaction;SMI could reverse the myocardial Ca<sup>2+</sup>overloading by resisting against lipid peroxidation and thus prevent adriamycin-induced myocardial injury.
