三种中药提取物对电刺激大鼠皮层惊厥阈值的影响及抗戊四唑惊厥作用 被引量:6

Anticonvulsant Effect of Three Traditional Chinese Herbal Medicines on Cortical Stimulation in Rats and Pentylene-Tetrazol Induced Convulsion in Mice

摘要 采用自动惊厥测定仪测定狼毒大戟、天南星、白蔹提取物的抗电刺激皮层惊厥阈值和抗戊四唑惊厥作用的结果。3种中药提取物以10ml/kg(相当于原生药1.2g/kg)腹腔注射。结果:给药后狼毒大戟组惊厥阈值提高(118±29)μA,效果持续10天;天南星组提高(119±22)μA,效果持续7天;白蔹组提高(83±19)μA,效果持续5天。抗戊四唑惊厥实验:对照组的惊厥潜伏期为3.5min,狠毒大戟为7.5min,天南星为7.4min,白蔹为6.8min。除给药当天动物有轻度镇静作用外,未见其他毒副作用。实验结果提示3种药提取物不但有明显的抗惊厥作用(P<0.001),而且药效持续较久。 The anticonvulsant effect of extracts of traditional Chinese herbal medicines have been tested in freely moving rats by deter- mination of threshold for localized seizures by means of direct cordeal stimulation via chronically implanted electrodes.Extracts of Euphorbia fischeriana,Rhizoma Arisaematis and Radix Ampelopis were administered intraperitoneally in a single dose of 10 mg/ kg.A control group received normal saline injection.The maximal increase in threshold compared to baseline values were 93± 29 uA(mean±SD),n=6)for Euphorbia fischeriana,119±22 uA for Rhizoma Arisaematis and 83±19uA for Radix Ampelop- is.These increases were significant(P&lt;0.01).Within 1 hour of the injection,the effect reached a plateau level which lasted 3 days for Radix Ampelopis,5 days for Rhizoma,na Arisaematis and 7 days for Euphorbia fischeriana.Apart from a slight sedative effect on the day of injection,no other side effects were observed.In the convulsion induced by pentylene-tetrazol incubation pe- riod of control group,Euphorvia fischeriana group,Rhizoma Arisarmafis group and Radix Ampelopis group were 3.5 min,7.5 min,7.4 min and 6.8 min,respectively.The results showed that the extracts of these three medicines exhibited not only dis- tinct effect on convulsive threshold,but also long duration of action.
