摘要
应用体外培养肺动脉平滑肌细胞(PASMC)并缺氧的方法,分别掺入~3H-TdR(胸腺嘧啶核苷)、~3H-脯氨酸,探讨中药制剂黄芪甙对缺氧培养的 PASMC 生长分泌的影响。细胞分常氧、缺氧两种方式培养,每种培养方式又分为黄芪甙组与正常对照组。无论缺氧与常氧,黄芪甙组与正常对照组相比,~3H-TdR 掺入值(反映 DNA 的合成即细胞增殖)都有显著下降,分别为常氧:每分钟液闪值(CPM 值)由2874.50±401.60下降到365.40±78.64,缺氧:CPM 值由5956.67±1977.49下降到1002.67±508.52(P【0.01)。黄芪甙同时抑制常氧、缺氧体外培养的 PASMC ~3H-脯氨酸掺入(反映胶原的合成、分泌),分别为常氧:CPM 值由3088.70±988.80下降到1977.60±911.97,缺氧:4714.93±1802.53下降到3180.50±1790.90(P【0.05),经两因素随机取样方差分析用药与缺氧两因素间交互影响显著。提示黄芪甙可抑制体外培养的PASMC 增殖及胶原分泌,黄芪甙尤其对缺氧引起的增殖分泌效应抑制作用更明显。
Cell culture technique was applied for detection of the effect of astragaloside in proliferation and collagen secretion of pul- rnonary artery smooth muscle cells (PASMC) by hypoxia.The incorporation of ~3H-TdR with the use of astragaloside decreased from 2874.5±401.6 to 365.4±78.64 in normal condition and from 5956.67±1977.49 to 1002.67±508.52 in hypoxia.The incorporation of ~3H-Prolin declined from 3088.7±988.8 to 1977,6±911.97 in normal condition,and from 4714.93±1802. 53 to 3180.5±1790.9 in hypoxia.It indicated that astragaloside could inhibit the proliferation and collagen secretion of PASMC in both hypoxia and normaxia,especially in hypoxia.
