摘要 目的 建立体外培养扩增树突状细胞 (DC)的方法 ,并利用DC和超抗原高聚金葡素 (HAS)诱导产生高效特异性抗肿瘤免疫。方法 用GM CSF和IL 4联合刺激诱导外周血单个核细胞分化为树突状细胞。HAS活化的淋巴细胞称为HASL。将DC与同源淋巴细胞或HASL共培养 ,分别称为DC L和DC HASL。用FCS分析细胞表型 ,MTT法测定细胞杀伤活性 ,并观察对裸鼠移植瘤的生长抑制作用。结果 在体外 ,DC HASL对GLC 82细胞具有相对特异的杀伤作用 ,HASL和DC L则无明显特异性。这三种效应细胞对荷瘤鼠的肿瘤生长都有不同程度的抑制 ,DC HASL抗瘤活性最强。结论 将DC和HAS结合可诱导产生一种高效并具有相对特异性的抗肿瘤免疫活性细胞。 Objective To culture dendritic cells(DC) in vitro and to induce specific anti-tumor immunoreaction in cancer cell lines with superantigen HAS and DC. Methods DC was cultured from PBMC by using GM-CSF and IL-4. Lymphocytes stimulated by HAS were named HASL. DC and lymphocyte or HASL were cocultured to induce specific anti-tumor effect, and the obtained cells were named DC-L or DC-HASL. Phenotypes of DC and HASL were analysed by FCS. MTT assay was used to determine cytotoxicity in vitro. The anti-tumor effects on GLC-82 bearing mice were also observed. Results DC-HASL could lyse GLC-82 cell specifically in vitro , but HASL and DC-L had no obvious specificity. These three effector cells can all inhibit the growth of tumor in nude mice, and the most effective one was DC-HASL. Conclusion High potency and relatively specific anti-tumor immunocyte can be prepared in virtue of DC and HAS.
