摘要
【目的】观察由黄连、生地、川乌、黄芪、党参、钩藤等 16味中药组成的解毒脱瘾汤对吗啡依赖大鼠镇痛及生殖功能的影响。【方法】采用大鼠戒断模型 ,观察解毒脱瘾汤对吗啡依赖大鼠下丘脑和血浆 β 内啡肽 (β EP)、结状神经节和孤束核P物质 (SP)以及血清内分泌激素的变化。【结果】 (1)吗啡依赖大鼠下丘脑 β EP的含量均较正常大鼠降低 ,解毒脱瘾汤高剂量组可增高下丘脑 β EP的含量 (P <0 0 1) ,解毒脱瘾汤中、高剂量组可增高血浆 β EP的含量 (P <0 0 1) ;吗啡依赖大鼠结状神经节和孤束核SP的含量升高 ,解毒脱瘾汤高剂量组可降低结状神经节和孤束核SP的含量 (P <0 0 1或P<0 0 5 )。 (2 )吗啡依赖雌性大鼠血清卵泡刺激素 (FSH)、雌二醇 (E2 )、泌乳素 (PRL)均低于正常对照组 ,解毒脱瘾汤中、高剂量组均可增高血清FSH、E2 、PRL的含量 (P <0 0 1或P <0 0 5 ) ;吗啡依赖大鼠睾丸酮 (T)含量显著降低 (P <0 0 1) ,解毒脱瘾汤中、高剂量组均可增高雄性大鼠血清T的含量 (P <0 0 1) ,而对大鼠血清黄体生成素 (LH)、PRL的含量无显著影响。【结论】解毒脱瘾汤可能通过改善吗啡依赖大鼠下丘脑—垂体—肾上腺的变化 ,消除其戒断症状 ,从而达到治疗的目的。
To observe the analgesic effect of Jiedu Tuoyin Decoction (JTD) and its influence on reproduction of morphine dependence rats. JTD is composed of Rhizoma Coptidis, Radix Rehmanniae, Radix Aconiti, Radix Astragali, Radix Codonopsis, Ramulus Uncariae cum Uncis, etc..Sixty rats were randomly allocated to six groups: normal saline group (Group A), morphine and normal saline group (Group B), morphine and clonidine group(Group C),morphine and low dosage of JTD group (Group D), morphine and moderate dosage of JTD group (Group E) and morphine and high dosage of JTD group (Group F).Content of β endophin(β EP) in hypothalamus and plasma, substance P (SP) content in ganglion nodosum(GN) and nucleus tractus solitarii(NTS) and serum levels of sexual hormones were examined.(1) β EP content in hypothalamus of morphine dependence rats was lower and SP content in ganglion nodosum and nucleus tractus solitarii was higher than that of normal rats (P<0 01);β EP content in hypothalamus of Group F and in plasma of Group E and Group F was increased and SP content in GN and NTS of Group F was decreased (P<0 05 or P<0 01 as compared with Group B). (2) Serum levels of follicle stimulating hormone (FSH), estradiol (E 2) and prolactin (PRL) in morphine dependence female rats and serum level of testosterone (T) in morphine dependence male rats was lower than that of normal rats (P<0 01); Serum levels of FSH, E 2 and PRL of female rats and serum T level of male rats in Group E and Group F was increased (P<0 05 or P<0 01 as compared with Group B) and the changes of serum levels of luteotrophic hormone(LH) and PRL in Group E and Group F stayed insignificant.[Conclusion]JTD can eliminate the withdrawal symptoms in morphine dependence rats probably by regulating the function of the axis of hypothalamus pituitary adrenal gland.
