摘要 目的 :用HPLC法测定人血浆中米安色林浓度。方法 :色谱柱为Hypersil ODS2 C1 8柱 ,流动相为甲醇 水 乙酸铵 (含 0 .0 5mol·L- 1 乙酸铵 ,0 .1mol·L- 1 氢氧化铵 ) =90∶7∶3,pH =7.7,UV检测器 ,波长为 2 4 0nm ,流速1mL·min- 1 。将待测血浆碱化后 ,重蒸无水乙醚分 2步提取 ,经含有标准参照品氯普噻吨的流动相溶解后测定。结果 :测定米安色林的线性范围为 5~ 14 0 μg·L- 1 (r =0 .9990 ) ,最低检测浓度为 5 μg·L- 1 ,日内RSD小于 5 .5 % ,日间RSD小于 8.8% ,方法回收率 >90 %。结论 :本方法简便、精密、准确 ,适用于米安色林的药代动力学研究 ,也可用于临床常规测定米安色林血浆浓度。 Objective:To establish a HPLC method for the determination of mianserin in human plasma.Methods:Chlorprothixene was used as the reference compound.The extracts of samples were chromatographed on a hypersil ODS 2 C 18 reversed phase column after two steps liquid liquid extraction by Diethyl ether absolute and monitored by UV detection at 240nm.The mobile phase consisted of CH 3OH∶H 2O∶ammonium acetate (containing of ammonium acetate and ammonia solution, 0.05 mol·L -1 and 0.1mol·L -1 respectively) (90∶7∶3,pH=7.7) at a flow rate of 1mL·min -1 .Results:The calibration curve was linear over the range of 5~140μg·L -1 ( r =0.9990).The intra day RSD was less than 5.5%, and the inter day RSD was less than 8.8%.The recovery was more than 90% and the limit of quantificaiton was 5μg·L -1 .Conclusion:The method has good selectivity, sensitivity and reproducibility and can be used for pharmacokinetic studies of mianserin in humans and the clinical determination of mianserin.
