摘要
目的 :观察血管内皮生长因子 (VEGF)对兔离体主动脉内皮细胞分泌功能的影响 ,了解一氧化氮 (NO)在其中所起的作用 ,以明确VEGF的作用机制。方法 :先进行兔主动脉内皮细胞的培养和鉴定 ,然后行传代培养。在第 3~ 5代主动脉内皮细胞培养液中加入不同浓度的VEGF ,培养 6h ,取上清液测定内皮素 (ET)、血管性血友病因子 (vWF)值 ,并应用一氧化氮合成酶 (NOS)抑制剂N -硝基 -L -精氨酸甲酯 (L -NAME)后观察ET、vWF值的变化情况。结果 :VEGF可抑制血管内皮细胞 (VEC)分泌ET、vWF ,并且呈剂量依赖性 (r =- 0 .96 881P <0 .0 1,r=- 0 .9711P <0 .0 1) ,NOS抑制剂L -NAME可剂量依赖性地阻断其效应 (P <0 .0 1)。结论 :NO在VEGF抑制VEC分泌ET、vWF中起中介作用 ,NO是VEGF作用机制中的一个重要通路。
Objective:To observe the effect of vascular endothelial growth factor (VEGF) on secretion of vascular endothelial cells of rabbit aorta in vitro, and to investigate the role of nitrogen monoxide in the process for elucidating the action mechanism of VEGF. Methods:The vascular endothelial cells of rabbit aorta were cultured and identified, then passage culture of the cells was carried out. The third- fifth generations of the vascular endothelial cells were cultured for 6 hours after putting different concentrations of VEGF into the culture medium.Endothelin (ET)?von Willebrand factor (vWF) in the upper solution of the culture medium were tested, and the changes of ET?vWF were observed after applying inhibitory agent-N-nitro-L-arginine methy ester (L-NAME) of nitrogen monoxide synthetase. Results:VEGF can inhibit secretion of ET?vWF of vascular endothelial cells dose-dependently?(r=-0.96881 P <0.01,r=-0.9711 P <0.01).L-NAME can inhibit the effect of VEGF dose-dependently( P <0.01). Conclusions:Nitrogen monoxide is an important mediator in the process of inhibiting the secretion of vascular endothelial cells by VEGF.Nitrogen monoxide is an important passage in the action mechanism of VEGF.
