摘要
目的 观察胚鼠皮质神经元遭受缺氧刺激时脑源性神经营养因子 (BDNF)对其的保护作用 ,并探讨该过程中酪氨酸激酶 B信使核糖核酸 (Trk Bm RNA)表达的变化。方法 对胚鼠皮质神经元进行体外培养 ,用流式细胞术检测不同缺氧时点实验组 (加入 BDNF)与对照组 (不加入 BDNF)的细胞活性差别 ;同时用原位杂交法检测 BDNF受体 Trk Bm RNA表达的改变。结果 BDNF对缺氧神经元有确切保护作用 ,加入 BDNF可引起Trk Bm RNA表达的上调。结论 BDNF可减轻神经元所遭受的缺氧损害 ,而 Trk Bm RNA表达上调可能是 BDNF发挥其对缺氧神经元保护作用的机理之一。
Objective To investigate the expression of brain derived neurotrophic factor (BDNF) receptor TrkBmRNA in hypoxia induced cortical neurons. Methods We compared the activities of the two groups of neurons with and without the BDNF in the culture fluids before exposed to the hypoxia conditions using flowing cytometry (FCM) and examined the changes of expressions of the BDNF receptor TrkBmRNA by in situ hybridization. Results Preincubating the neurons with BDNF for 24 hours or adding BDNF just before exposing to the hypoxic conditions protected the cortical neurons from hypoxia induced neurotoxicity. The possible mechanism could be the up regulation of the transcription of the TrkBmRNA. Conclusion BDNF plays an important role in preventing cortical neurons from hypoxia induced neurotoxicity, and the up regulation of TrkBmRNA is possibly one of the mechanisms of the survival.
