摘要
目的 检测细胞周期调节蛋白 P16、CDK4和 P5 3在成人和儿童髓母细胞瘤中的表达 ,并探讨其与预后的关系。方法 应用免疫组化 ABC法对 2 1例成人和 14例儿童髓母细胞瘤石蜡切片进行免疫组化检测 ,并结合随访资料进行生存曲线分析。结果 在全部 35例标本中 ,P16阳性率为 6 2 .9% ,CDK4阳性率为 10 0 % ,P5 3阳性率为 2 8.6 % ,其中 P16蛋白在成人中的表达 (16 / 2 1)要高于在儿童中的表达 (6 / 14 ) ,P5 3蛋白在成人中的表达(3/ 2 1)要低于在儿童中的表达 (7/ 14 ) ,但上述 3种蛋白的表达均与预后无关。结论 P16蛋白的表达缺失和 P5 3蛋白的异常表达可能与髓母细胞瘤的发生有关 ,但对预后无意义。儿童髓母细胞瘤的遗传学异常现象高于成人。
Objective To detect the expression of P16, cyclin dependent kinase 4 and P53 in adult and childhood medulloblastomas and the possible prognostic value thereof. Methods ABC immunohistochemistry was used to detect the expression of P16, CDK4 and P53 in the paraffin embedded medulloblastoma specimens of 21 adults and 14 children. Follow up data were obtained to perform the Kaplan Meier survival analysis. Results In the 35 cases, positive expression ratios of P16, CDK4 and P53 were 62.9%, 100% and 28.6%, respectively. The positive expression ratio of P16 in adult cases(16/21) was higher than that in childhood cases(6/14), whereas the positive expression ratio of P53 in adult cases(3/21) was lower than that in childhood cases(7/14). The expressions of P16, CDK4 and P53 had no correlation with the prognosis by Kaplan Meier survival analysis. Conclusion Deletion of P16 expression and abnormal expression of P53 might contribute to the tumorigenesis of medulloblastomas. This is apparent in childhood cases, though no prognostic value has been noted in this study. [
