雾化吸入γ-干扰素对免疫低下大鼠肺部抗感染能力的增强作用 被引量:2

摘要 目的 研究雾化吸入 γ-干扰素 (IFNγ)对免疫低下大鼠肺部抗感染能力的影响。方法 检测气管内注射白色念珠菌、或气管内注射白色念珠菌并雾化吸入 IFNγ1、3、7d的免疫低下大鼠肺泡巨噬细胞 (AM)的吞噬及杀菌功能 ,抗原提呈功能 ,培养上清中 TNF- α活性和 IL- 1β、IL- 6水平 ,以及血淋巴细胞杀伤功能和血清 IFNγ活性 ,IL- 1β、IL- 6水平 ,并于第 7天进行左肺白色念珠菌培养计数。结果 雾化吸入 IFNγ组大鼠肺白色念珠菌计数显著少于对照组大鼠 (P<0 .0 5 ) ;雾化吸入 IFNγ组大鼠 AM的吞噬及杀菌百分率、Ia抗原表达的阳性率、AM培养上清中 TNF- α的活性及 IL- 1β和 IL- 6水平显著高于对照组大鼠 (P<0 .0 1,P<0 .0 5 ) ;两组间血清 IFNγ活性、IL- 1β和 IL- 6水平 ,以及感染后第 7天淋巴细胞杀伤功能差异无显著性 (P>0 .0 5 )。结论 雾化吸入 IFNγ可明显增强免疫低下大鼠肺部细胞免疫反应和抗感染能力 ,但对血细胞免疫状态无明显影响。 OBJECTIVE To study the influence of IFNγ via aerosol on the anti infection ability in the lungs of the immunocompromised rats. METHODS The phagocyting and bactericidal function, the Ia antigen expression, the TNF α activity and the levels of IL 1β and IL 6 in the culture supernate of the alveolar macrophage(AM), the IFNγ activity and the levels of IL 1β and IL 6 in the serum, and the killing ability of the lymphocytes in the immunocompromised rats administrated IFNγ via aerosol and the control groups were investigated when the 1, 3, 7 days after they were tracheally injected Candida albicans. And the C. albicans count of the left lung was investigated at the 7th day after. RESULTS The C. albicans count of the left lung of the immunocompromised rats administrated IFNγ via aerosol was significantly less than that of the control group(P<0.05). The phagocyting and bactericidal percentages, the Ia antigen expression percentages, the TNF α activity and the levels of IL 1β and IL 6 in the culture supernate of the AM of the immunocompromised rats administrated IFNγ via aerosol were markedly higher than those of the control groups (P<0.01, P<0.05). The IFNγ activity and the levels of IL 1β and IL 6 in the serum, and the killing ability of the lymphocytes in the blood of the immunocompromised rats administrated IFNγ via aerosol had no difference with those of the control groups (P>0.05). CONCLUSIONS Administration of IFNγ via aerosol can obviously enhance the cellular immunity and anti infection ability of the lungs, but has no influence on the whole body cellular immunity of the immunocompromised rats.
