摘要 目的 探讨某火力发电厂粉煤灰对染尘早期大鼠肺组织的损害作用及其对细胞因子释放的影响。方法 Wistar大鼠一次性气管内注入染尘 1ml(5 0mg/ml) ,2 ,4 -二硝基苯肼比色法、Folin -酚试剂法和钼蓝法测定支气管肺泡灌洗液 (BALF)中乳酸脱氢酶活力及总蛋白和磷脂含量 ,免疫组织化学染色法直接检测肺组织中的肿瘤坏死因子 -α (TNF α)和转化生长因子 -α (TGF α)的表达。结果 与生理盐水组相比 ,石英组和粉煤灰组大鼠BALF中乳酸脱氢酶活力增高 ,总蛋白和磷脂含量明显增加 (P <0 0 1) ,石英组增加幅度大于粉煤灰组 (P <0 0 1)。生理盐水组未见TNF α表达 ,石英和粉煤灰均引起TNF α表达 ,每高倍镜视野下 (× 4 0 0 )两组阳性细胞百分数分别为 (41 2 4± 3 2 6 ) % ,(30 33± 2 86 ) % ,差异有显著性(P <0 0 1)。生理盐水组和粉煤灰组肺泡上皮细胞TGF α表达分别为 (2 19± 0 2 2 ) %和 (2 4 9± 0 2 9) % ,P >0 0 5 ,而石英组TGF α表达明显增多 ,为 (79 5 7± 5 0 9) % ,P <0 0 1。结论 粉煤灰能够引起染尘早期大鼠肺组织损害和TNF α表达增强 。 ObjectiveTo explore the toxicity of fly ash from th e thermal power station on rat lung tissues,and the role of fly ash in the release of cytokines. MethodsWistar rats were exposed to quartz and fly ash( 50 mg/ml) BFQ respectively by single tracheal instillation.The activity of lactate dehydrogenase,total protein and phospholipid contents were determined in bronchoalveolar lavage fluid(BALF). BFQ Tumor necrosis factor-α B F (TNF-α) BFQ and transforming growth factor-α(TGF-α BFQ )in lung tissues were determined by -histochemistry. JP ResultsIn comparison with control groups,the activity of l acta te dehydrogenase,total protein and phospholipid contents were significantly elev ated in groups exposed to fly ash( WTBX P<0.01), BFQ while i n qu artz exposed groups,they were even higher than those of the fly ash exposed grou ps( WTBX P<0.01). BFQ JP2 The lung tissues exposed to JP quartz and fly ash had highly significant increase of TNF-α immuno\|staining[(41.24 ±3 .26)%and(30.33±2.86)%], BFQ however,the unexposed animals exhibited no TN F-α labeling.The pulmonary epithelia of control rats showed little TGF-α imm uno-staining[(2.19±0.22)% BFQ ].The fly ash did not cause the incr ease in TGF-α expression[(2.49±0.29)%].A significant i ncrease of the TGF-α expression was observed in the lung tissue cells of rats exposed to quartz[(79.57±5.09)%, WTBX P<0.01 BFQ ]. ConclusionsIn the early exposure stage,the fly ash could induc e the injury of lung tissues of rats and the enhancement of TNF-α release,but the potency of fly ash was lower than that of quartz.
