摘要
目的 研究脂肪酶催化壳聚糖降解的特性。 方法 用粘度法测定溶液粘度和分光光度法还原糖浓度作为判断依据 ,研究各种因素对脂肪酶催化壳聚糖降解的影响。 结果 确定以壳聚糖为底物的脂肪酶的催化的特征 :最适宜温度为 4 0~ 5 0℃ ,最适 p H值为 3左右 ,反应不遵循 Michaelis- Menten动力学方程 ,壳聚糖脱乙酰度在 70 %~ 90 %降解速度较快。 结论 在弱酸性条件下 ,脂肪酶能非常有效地降解壳聚糖 ,该反应条件温和 ,速度快 。
Objective\ To research on the depolymerization of chitosan with the aid of lipase.\ Methods\ By determinating the viscosity and concentration of reducing sugar in chitosan solution, the effects on the efficiency of catalytic degradation of chitosan by lipase were investigated.\ Results\ The data revealed that the enzymatic process did not obey Michaelis\|Menten kinetic model.\ The optimal condition enzymatic hydrolysis of chitosan were: pH 3.0, temperature 40 ~50 ℃, deacetylation degree of chitosan is 70%~90%.\ Conclusion\ The preparation of water\|soluble oligosaccharides of chitosan by catalytic degradation with lipase is practicable.\;
