余姚白蒜组织培养脱毒的研究 被引量:3

摘要 以商用余姚白蒜(Allium sativum L.)等休眠鳞茎蒜瓣蒂上切取的生长点等为试验材料,以MS、B_5和Nitsch(1969)为基本培养基,在其上分别添加萘乙酸(NAA)、吲哚乙酸(IAA)和2,4-二氯苯氧乙酸(2,4-D)、6-苄基腺嘌呤(6-BA)、玉米素(ZT)和细胞激动素(KT)以及赤霉素(GA_3)等生长调节剂进行培养。结果以MS和B5两种培养基诱导产生愈伤组织的效果较好。诱导分化成苗的生长调节剂为NAA,浓度范围在0.1-0 5mg·L^(-1)内,其累计分化成苗率达80.1%,其次是6-BA和IAA,浓度范围分别为0.2-0.5mg·L^(-1)和0.2-1.0mg·L^(-1),其累计分化成苗率均为53.6%。成苗株的脱毒率达100%。 Segments dissected from the apex of dormant bulblet of Yuyao White Garic (Allium sativum L.) were cultured on the basal medium of MS or B5 or Nitsch , supplemented with NAA ,IAA, 2, 4-D. 6-BA, ZT, KT, and GA_3.The results showed that both the MS and B_5 were favourable for callus formation.In growth regulators used in the experiment NAA, 6-BA and IAA could induce to develop plantlets in high propotion , the total rate of plantlet developed (TRPD) for NAA reached to 80.1% when its concentrations were in the range of 0.1-0.5mg.L^(-1), for 6-BA and IAA their TRPD both reached to 53.6% at the concentrations of 0.2-0.5mg.L^(-1)and 0.2-1.0mg鈥?L^(-1), respectively .The rate of virusfree garlic produced by tissue culture in the study was 100%.
