球毛壳菌原生质体形成与再生研究 被引量:3

摘要 采用酶解法制备球毛壳原生质体 .确定酶解条件为 :菌龄 2 .5d ,渗稳剂为S/C .采用 5、15、15mg/mL的Novozym 2 34、蜗牛酶 ,溶壁酶混合酶液 ,30℃下酶解 1.5h .再生率可达 2 9.11% .研究了渗稳剂对原生质体制备与再生的影响 ,并对球毛壳原生质体的释放和再生过程进行了观察 .结果表明 :山梨醇可促进原生质体的释放和再生 ;Ca2 + 对原生质体的再生有促进作用 ;葡萄糖对再生菌株生长的促进作用优于山梨醇 . Protoplasts of Chaetomium globosum were obtained successfully by using a mixed enzyme solution of 5 mg/mL of Novozym 234, 15 mg/mL of snailase and 15 mg/mL of lywallzym. The mycelia of 2.5 d old could be enzyme digested in 1.5 h at 30 ℃.The regeneration rate of protoplasts went up to 29% . Sorbital could improve the formation and regeneration of protoplasts while Ca 2+ improved regeneration only. Glucose improved the growth of regeneration colonies more effectively than sorbital. The protoplasts releasing and regenerating of Chaetomium globosum were observed during this study.
