几种红花黄酮醇体外抑制PAF诱导PMN聚集及黏附作用 被引量:1

摘要 目的 研究红花成分槲皮素、山奈酚、杨梅酮、芦丁等黄酮醇对血小板激活因子 (PAF)诱导的家兔多型核白细胞 (PMN)聚集和黏附作用的影响。方法 明胶沉降法制备PMN ;比浊法观察PAF诱导的PMN聚集 ;用比色法测量溶液吸光度表示黏附活性。结果 黄酮醇可明显抑制PAF诱导的PMN聚集及黏附活性的升高 (P均 <0 .0 5 ) ,其中槲皮素、山奈酚、杨梅酮、芦丁抑制PAF诱导PMN聚集的IC50 分别为 0 .72 8,0 .5 4 2 ,0 .10 4 ,4 .4 5mmol·L-1。 OBJECTIVE: The inhibitory effects of quercetin, kaempferol, myricetin and rutin from Carthamus tinctorious L. against rabbit polymorphonuclear leucocyte (PMN) aggregation and adhesion induced by platelet activating factor(PAF) were studied. METHOD: Rabbit PMN was prepared through gelatin precipitation procedure. After induced by PAF, PMN aggregation was observed by turbidity assay. Coloured PMN, which was activated by PAF and adhered to the surface of plastics, was broken and the A570 nm of the solution was measured to show the adhesion potency. RESULTS: The aggregation rate and A570 nm were decreased evidently (P 50 of quercetin, kaempferol, myricetin and rutin in aggregation test were 0.728, 0.542, 0.104 and 4.45 mmol&middotL-1 respectively. CONCLUSION: It was suggested that the inhibitory effects against PMN aggregation and adhesion induced by PAF of these flavonols were dose-dependent.
