细胞周期素H和细胞周期素依赖性激酶7在人乳腺癌组织中的表达及其意义 被引量:4

A study on the expression of cyclin H and CDK 7 in human breast cancer

摘要 目的研究人乳腺癌组织中细胞周期素H(cyclinH)和细胞周期素依赖性激酶 7(CDK7)蛋白表达以及术前化疗对其表达的影响。方法应用固定化蛋白质印迹法检测 40例乳腺癌及相应正常乳腺组织cyclinH和CDK7蛋白表达。结果乳腺癌组织cyclinH和CDK7表达水平明显高于正常乳腺组织 (t值分别为 9 5 2 4和 2 984,P <0 0 1)。Ⅲ期乳腺癌与Ⅰ、Ⅱ期乳腺癌相比 ,差异有显著性 (t值分别为 6 0 3和 4 6 9,P <0 0 5 ) ;直径大于 5cm的乳腺癌cyclinH表达水平高于直径小于 2cm者 (t=4 49,P <0 0 5 )。术前行化疗的乳腺癌cyclinH蛋白表达水平明显下降 (t =6 .5 2 ,P <0 0 5 )。结论 cyclinH和CDK7在乳腺癌中呈过度表达 。 ObjectiveTo investigate the expression of cyclin H and cyclin-dependent kinase 7(CDK7)in human breast cancer and the effect of preoperative chemotherapy on their expression.MethodsWestern blotting was used to measure the expression of cyclin H and CDK7 proteins in breast carcinoma and adjacent normal tissue in 40 patients. Results cyclin H and CDK7 protein levels elevated in breast cancer tissues in comparison with adjacent normal tissues (P<0 01). Protein levels of cyclin H and CDK7 were higher in stage Ⅲ tumors than that in stage Ⅰ and stage Ⅱ (P<0 05). In the 11 patients receiving preoperative chemotherapy, tumor cyclin H and CDK7 protein levels significantly decreased.Conclusion cyclin H and CDK7 may play an important role in the development of breast cancer.Preoperative chemotherapy inhibited cyclin H and CDK7 expression in tumor.
