术前使用FTY720诱导小鼠对同种异体心脏移植物的耐受 被引量:1

摘要 目的 观察术前使用FTY72 0对小鼠同种异体心脏移植物存活时间的影响。方法 以小鼠颈部异位心脏移植为模型 ,供者为BALB/c小鼠 ,受者为C57BL/ 6小鼠。受鼠分为 4组 :A组为空白对照组 ;B组术前 3d至术后 1 1d每日管饲FTY72 0 ;C组手术当天至术后 1 4d每日管饲FTY72 0 ;D组仅在术前 3d至手术当天每日管饲FTY72 0。FTY72 0的剂量都为 3mg·kg- 1 ·d- 1 。结果 A组移植物中位存活时间 (MST)为 8d ;B组所有移植物存活时间均超过 2 7d ,有半数 (6/ 1 1 )移植物存活超过1 0 0d ;C组移植物存活时间显著延长 (MST =1 6d) ;D组移植物中位存活时间为 1 4d ,并有 1只超过1 0 0d。结论 术前使用FTY72 0可以延长同种异体心脏移植物的存活时间 。 Objective To observe the effect pretreatment with FTY720 on the survival time of mouse heart allograft. Methods Eight to ten weeks old male C57BL/6 mice were randomly divided into four groups and the male BALB/c mice were chosen as donors. Group 1 was the nil treated control. Groups 2,3 and 4 were given a monotherapeutical course of FTY720 at the dose of 3?mg/kg every day by oral gavage . The treatment of group 2 was started 3 days before the operation and stopped at day 11 (15 doses in total ). Group 3 started at the ordinary day 0 and stopped at day 14 (also 15 doses in total). Group 4 started 3 days before and stopped at day 0. Results The survival time of all grafts in the FTY720 treated groups was prolonged as compared with the controls (7~8 days, n =6). Whereas the recipients in the group 3 rejected their allografts at a median survival time (MST) of 16 days ( n =6),those in the group 2 showed significant prolongation of allograft survival (all> 27?d , n =11) and 6 of them exceeded 100 days. The MST of group 4 was 14 days ( n =5),and one case exceeded 100 days.Conclusion Pre operative use of FTY720 can prolong the survival time of the mouse heart allograft and induce the tolerance of recipients to mouse heart allograft.
