转化生长因子β_1反义寡核苷酸抑制单侧肾切除糖尿病大鼠系膜增生 被引量:3

Antisense transforming growth factor-β_1 oligonucleotide inhibits mesangial hyperplasia in unilateral nephrectomic diabetic rats

摘要 目的 探讨转化生长因子 (TGF) β1反义寡核苷酸对单侧肾切除糖尿病大鼠系膜增生的作用。方法 采用人工合成TGF β1反义、正义及错配的 3组寡核苷酸经脂质体包裹后转染单侧肾切除糖尿病大鼠肾小球系膜细胞 (MC)。用细胞计数检测转染后MC的生长抑制率 ,用RT PCR和ELISA方法检测TGF β1mRNA和蛋白的表达水平 ,并检测Ⅳ型胶原mRNA表达水平的变化。结果TGF β1反义寡核苷酸可抑制MC的存活 ,TGF β1mRNA及蛋白的表达水平下降 ,Ⅳ型胶原mRNA的水平也下降。结论 TGF β1反义寡核苷酸不仅抑制TGF β1mRNA及蛋白的表达 ,而且还可抑制细胞外基质Ⅳ型胶原mRNA的表达 ,抑制MC的肥大 ,从而抑制单侧肾切除糖尿病大鼠系膜的增生。 Objective To investigate the inhibitory effects of antisense transforming growth factor (TGF) β 1oligonucleotideonthe mesangial hyperplasia in unilateral nephrectomic diabetic rats. Methods TGF β 1 antisense, sense and missense oligonucleotides were synthesized and transfected to mesangial cells (MC) from diabetic rats with unilateral nephrectomy by lipofectamine. Their effects on the TGF β 1 gene and the collagen Ⅳ gene were studied by RT PCR while the expression of TGF β 1 protein was measured by ELISA. Cell count technique was also used to evaluate the MC survival. Results The MC survival could be inhibited by TGF β 1 antisense oligonucleotide. TGF β 1 antisense oligonucleotide inhibited TGF β 1 expressions at both mRNA and protein levels and inhibited the expression of collagen Ⅳ mRNA as well. Conclusion The MC survival is inhibited by TGF β 1 antisense oligonucleotide. TGF β 1 and collagen Ⅳ production as well as the expression of extracellular matrix are also inhibited by its expression, which suggests that the mesangial hyperplasia in unilateral nephrectomic diabetic rats can be prevented by TGF β 1 antisense oligonucleotide.
