摘要
①目的 探讨 β 连环蛋白 (β cat)的异常表达和细胞周期素D1 (cyclinD1 )高表达与胰腺癌发生、浸润、转移和预后的关系。②方法 应用免疫组织化学PicTureTM 二步法 ,检测 47例胰腺癌组织、1 2例胰腺导管上皮内肿瘤 (上皮中度不典型增生 )和 1 0例正常胰腺组织中 β cat和cyclinD1的表达。 ③结果 β cat在 1 0例正常胰腺组织均呈正常表达 ,而cyclinD1均呈阴性。β cat在胰腺导管上皮内肿瘤和胰腺癌中的表达率分别为 50 .0 %和 68.1 % ,两者比较差异无显著性 (χ2 =0 .689,P >0 .0 5) ;cyclinD1在胰腺导管上皮内肿瘤和胰腺癌中的表达率分别为 50 .0 %和 74.5 % ,两者比较差异无显著性 (χ2 =0 .2 1 3 ,P >0 .0 5)。β cat异常表达率与胰腺癌的转移和 1年生存率显著相关 (χ2 =6 .2 30 ,5 .495 ,P均 <0 .0 5)。cyclinD1的表达率与胰腺癌的增殖和分化程度有关 (P =0 .0 0 6 ,χ2 =6 .1 90 ,P <0 .0 5)。β cat异常表达与cyclinD1高表达在胰腺导管上皮内肿瘤和胰腺癌中均呈显著的正相关 (P =0 .0 0 2 ,χ2=6 .937,P <0 .0 1 ;r=1 .0 0 0 ,0 .845 ,P <0 .0 1 )。④结论 β cat异常表达导致cyclinD1的激活而引起细胞增殖和分化失控 ,在胰腺癌的发生机制中可能发挥重要作用。β
Objective\ To study the relationship between the abnormal expression of β catenin (β cat) and the high expression of cyclin D1(D1) and the occurrence, infiltration, metastasis and prognosis of pancreatic cancer.\ Methods\ Immunohistochemical PicTure TM was used to test the expression of β cat and D1 in 47 cases of pancreatic cancer , 12 cases of the pancreatic intraepithelial neoplasm and 10 cases of normal tissue of the pancreas.\ Results\ β cat was expressed normally in the 10 cases of normal pancreatic tissue, while D1 was negative. The expression rates of β cat in the tissues of pancreatic intraepithelial neoplasms and pancreatic cancer was 50.0% and 68.1%, and D1 was 50.0% and 74.5%, respectively. No significant difference was found ( χ 2= 0.213, P >0.05). The abnormal expression rate of β cat was significantly correlated with the metastasis of the pancreatic cancer and its 1 year survival ( χ 2=6.230, 5.495, P <0.05). The expression rate of D1 was correlated with the proliferation of the pancreatic cancer and extent of differentiation ( χ 2=6.190, P<0.05; P =0.006). The abnormal expression of β cat and high expression of D1 had a parallel relationship with the pancreatic intraepithelial neoplasms and pancreatic cancer ( P=0.002, χ 2= 6.937, P< 0.01; r=1.000, 0.845, P <0.01 ).\ Conclusion\ The abnormal expression of β cat activates D1, and results in unchecked proliferation and differentiation of the cell, which probably play an important role in the genesis of pancreatic cancer. The abnormal expression of β cat can be used as a molecular biological index to determine the metastasis and prognosis of the disease.
