摘要
①目的 探讨系统性红斑狼疮 (SLE)儿童外周血淋巴细胞凋亡特性及其与病情和部分免疫指标的相关性。②方法 应用形态法、间接免疫荧光法和免疫金斑法 ,检测 1 8例SLE病儿和 1 5名健康儿童外周血淋巴细胞凋亡率、T淋巴细胞亚群、CD+2 5细胞百分率和抗ds DNA抗体。③结果 培养前及培养 48h ,SLE病儿组外周血淋巴细胞凋亡率均显著低于正常对照组 (t=2 .363 ,2 .954 ,P <0 .0 5 ,0 .0 1 ) ;培养 48h ,活动期病儿外周血淋巴细胞凋亡率明显低于稳定期病儿 (t=2 .2 94,P <0 .0 5) ,抗ds DNA抗体阳性组外周血淋巴细胞凋亡率显著低于抗ds DNA抗体阴性组 (t=2 .92 8,P <0 .0 1 ) ,SLE病儿外周血淋巴细胞凋亡率与CD+4 细胞百分率、CD+4 CD+8细胞比值和CD+2 5细胞百分率皆呈负相关 (r=- 0 .49~ - 0 .59,P <0 .0 1 )。④结论 SLE病儿外周血淋巴细胞凋亡减少 。
Objective\ To study the features of apoptosis of peripheral blood lymphocytes and its relationship with the disease and some immune parameters in children with systemic lupus erythematosus(SLE). \ Methods\ The susceptibility to apoptosis of lymphocytes was determined in 18 children with SLE and 15 normal children by percentage calculation of apoptosis cells. The T lymphocyte subsets, percentage of CD + 25 cells, and serum anti ds DNA antibody were determined by immunofluoscence and colloidal gold immune procedure. \ Results\ The apoptosis cell percentage of lymphocytes incubated for 0 and 48 h in children with SLE were all significantly lower than that in normal children ( t=2.363, 2.954,P <0.05, 0.01), and in active stage of illness significantly lower than in steady stage ( t=2.294, P < 0.05). The apoptosis rate for serum anti ds DNA antibody group was lower than for non anti ds DNA antibody ( t= 2.928, P <0.01). The apoptosis cell percentage of lymphocytes had negative correlation with the percentage of CD + 4 cells, ratio of CD + 4 /CD + 8 cells and percentage of CD + 25 cells ( r=-0.49--0.59, P <0.01) in children with SLE. \ Conclusion Reduced susceptibility to apoptosis of peripheral blood lymphocytes is found in children with SLE, which is closely related to the functional disorder of cell or body fluid mediated immunity in children with SLE.
