摘要
目的 :探讨静脉输注TNF α基因修饰的舌癌DNL在舌癌动物模型体内的分布规律。方法 :用氚标胸腺嘧啶核苷 (3 HTdR)标记TNF α基因修饰的舌癌DNL ,经尾静脉输入人舌癌 (Tca8113细胞 )原位移植模型体内 ,分别检测注射后 5min、6、12、2 4、48、168h肿瘤组织及各脏器单位质量的放射活性 (衰变率 ,DPM )。结果 :输注后 5minDNL细胞大部分见于肺 ,6h单位质量DPM值脾最高 ,其次是肝、肺 ;2 4h由高到低依次是 :脾、肺、肝 ;12h瘤体单位质量DPM值是外周血的 3~ 4倍。结论 :人舌癌DNL可能对人舌癌病灶有一定的靶向聚集作用 ,是基因治疗较为理想的效应细胞。
Objective: To investigate the distribution of TNF α gene transduced drainage node of lymphocytes( DNL) obtained from human tongue cancer in nude mice bearing human tongue carcinoma. Methods: 3HTdR labeled DNLs modified with TNF α gene were transferred in the murine tumor models by intravenous infusion, and the relative radioactivity (DPM/g) in tumor and normal tissues was detected after 5 min, 6 h ,12 h, 24 h,48 h and 168 h.Results: After intravenous infusion 3H labeled DNLs were the most in lung in 5 min,in spleen 6 h, in tumor from 12 to 168 h respectively. Conclusion: DNLs from human tongue cancer can infiltrate into tumor tisse, and may be suitable target cells for gene therapy.
