摘要
目的 :研究机械张力对成骨细胞增殖能力的影响。方法 :通过Flexercell细胞拉伸力学装置对人成骨样细胞MG 63施加 6%、12 %和 2 4%拉伸应变加载实验 ,用MTT法检测细胞受力后的增殖变化。结果 :细胞受不同大小张力作用后 ,其增殖能力均增强 ,但以 12 %拉伸应变率的促增殖作用最为显著。结论 :机械张力可以促进成骨细胞的增殖能力 。
Objective: To study the effects of mechanical strain on the proliferation of human osteoblasts. Methods: Cloned MG 63 human osteoblasts were cultured and seeded at 10 5/well in 6 well Bioflex cell cuture plates. Subsequently, the cells were exposed to mechanical strain stimulation (at 6%, 12% and 24% elongation rate respectively) for 24 hours using Flexercell 3000 Cell Stretching Unit. MTT colorimetric method was used to assess the proliferation of the cells. Results: The proliferation of MG 63 cells was significantly increased 24 h after mechanical strain treatment( P <0.05), 12% stretching showed the strongest stimulatory effects ( P <0.05). Conclusion: Mechanical strain can stimulate the proliferation of osteoblasts. The effect might be related to the magnitude of mechanical loading.
