不同预处理方法建立幽门螺杆菌感染动物模型的比较 被引量:3

Establishment of Animal Models of Helicobacter pylori Infection by Various Pretreatments

摘要 目的 确定感染率高且稳定的幽门螺杆菌感染动物模型预处理方案。方法 取蒙古沙鼠200只,随机分为3个实验组及1个对照组。实验组沙鼠在断食、水12h后分别应用pH2盐酸、消炎痛+胃复安和50%乙醇3种不同方案进行预处理,对照组用生理盐水处理。此后继续断食、水12h,再灌喂Hp菌液(109cfu/ml)0.5ml/只。共3次,每次间隔12h。最后1次灌喂后2h给食、水。后每隔4周解剖一批动物,每组10只,进行分离培养、ELISA、PCR、快速尿素酶检测和病理切片检查。结果3组实验动物感染率不同,其中50%乙醇组沙鼠感染率相对较高,达到80%,其病理组织学变化与人体相应病变也极为相似。结论50%乙醇组沙鼠预处理方案感染效果最好,可作为Hp感染动物模型的预处理方法。 Objective To compare the effects of various pretreatments on the infection of Helicobacter pylori in Mongolian gerbils and develop a method for the establishment of a suitable animal model with high infection rate. Methods A total of 200 Mongolian gerbils were divided randomly into one control and 3 experimental groups. After being fasted for 12h, the animals in 3 experimental groups were pretreated with hydrochoric acid(pH 2),confortid plus metoclopramide and 50% alcohol respectively, and those in control group with physiological saline. Each of animals was further fasted for 12h and orally administered with 0.5ml of Hp liquid (109 cufu/ml) for 3 times each at an interval of 12h and fed with foods and water 2h after the last administration . Ten animals of each group were dissected every 4 weeks and detected for serum Hp antibody by ELISA. The gastric mucosa was isolated for detecting Hp by rapid urease test and PCR, as well as bacterial culture and pathological examination. Results The infection rate of the animals pretreated with 50% alcohol was the highest (80%) in the 3 experimental groups,and the histopathological changes of them were also similar to those of human beings. Conclusion 50% alcohol is suitable for the pretreatment of animal model of Hp infection.
