摘要 目的 : 检测蕈样肉芽肿 (MF)的端粒酶活性和细胞增殖情况 ,探讨它们在MF肿瘤发生机制中的作用及关系。方法 : 使用免疫组化法检测增殖细胞核抗原 (PCNA)的表达情况 ;聚合酶链反应—酶联免疫吸附测定法检测端粒酶活性。结果 : 肿瘤期MF、斑块期MF、斑片期MF、扁平苔藓和正常皮肤的PCNA细胞增殖指数分别为 2 2 .8、12 .1、8.8、1.7和 0 ;端粒酶活性的A值分别为 0 .737、0 .313、0 .2 4 0、0 .0 4 5和 0 .0 4 4。端粒酶阳性的MF的PCNA细胞增殖指数高于端粒酶阴性的MF。结论 : 端粒酶活性存在于大部分MF标本中 ,端粒酶活性与MF的恶性程度及分期有关。端粒酶活性与细胞增殖指数有关 。 Objective: To detect the telomerase activity and cell proliferation in patients with mycosis fungoides (MF) and to explore their roles in tumor genesis and relationship in MF. Method: The expression of PCNA was tested with immunochemistry and telomerase activity was measured with polymerase chain reaction-enzyme linked immunosorbent assay (PCR-ELISA).Results: PCNA cell proliferation indexes were 22.8, 12.1, 8.8, 1.7 and 0 in tumor, plaque and patch stages of MF, lichen planus and normal skin, respectively. The values of telomerase activity were 0.7373, 0.313, 0.240, 0.045 and 0.044 in tumor, plaque and patch stages of MF, lichen planus and normal skin, respectively. PCNA index was higher in MF with positive telomerase than that with negative telomerase activity. Conclusion : Telomerase activity was positive in the most of patients with MF, correlating with the malignant grade and stage of MF. Telomerase activity was correlated with cell proliferation index and the activation of telomerase was prerequisite for infinite proliferation of tumor cells.
