摘要
目的 :对真菌 2 2 0 5的代谢产物进行活性跟踪研究 ,分离出具有拮抗白细胞介素 1受体活性的小分子化合物 2 2 0 5A。方法 :采用大孔吸附、溶媒萃取、硅胶柱色谱、ODS反相柱色谱方法进行分离和纯化 ,以酶联免疫法进行活性跟踪 ,通过UV ,IR ,FABMS ,NMR和元素分析进行化合物的结构鉴定。结果 :分离得到的化合物 2 2 0 5A的相对分子质量为 2 80 ,根据波谱解析 ,确定其为已知化合物布雷菲德菌素A。结论 :通过白细胞介素 1受体阻断剂筛选模型筛选出活性物质布雷菲德菌素A 。
Objective:To study the interesting compound 2205A from the metabolites of fungus sp.of 2205.Methods:The compound was isolated by adsorption with X5 resin, extraction with dichloromethane, silica gel column chromatography and purified by ODS reverse column chromatography.The activity was tested with ELISA.And then the compound was identified through element analysis,UV,IR,FABMS and NMR.Results:The molecular weight of 2205A is 280.According to the spectroscopy study,it was identified as a known compound brefeldin A.Conclusion:It is firstly reported that brefeldin A was screened as an antagonist of interleukin 1 receptor.
