睾酮在体外对脾淋巴细胞增殖反应和分泌IgG的影响 被引量:5

摘要 为探讨雄激素对大鼠脾淋巴细胞产生免疫球蛋白和增殖反应的影响 ,用不同浓度睾酮作用于脾淋巴细胞 ,用中性红活染比色法和四甲基偶氮唑兰法 (MTT)分别观察睾酮对培养的大鼠脾淋巴细胞增殖的影响 ;用酶联免疫法 (ELISA)检测不同浓度睾酮处理的脾淋巴细胞由刀豆素A(ConA)刺激产生白细胞介素 6(IL- 6)和免疫球蛋白 (IgG)的影响。结果发现在睾酮作用下 ,细胞增殖反应下降 ;睾酮可下调脾淋巴细胞产生的IgG。 The purpose of the present study was to investigate the effects of testosterone on proliferative response and IgG production by splenic lymphocytes in vitro. Splenic lymphocytes from rats were cultured in the presence of the indicated doses of testosterone propionate for 72 h. The cell viability was assessed by a trypan blue exclusion test. Proliferation was determined by3 (4,5 dimethylthiazol 2 yl) 2,5 diphenyltetrazolium bromide (MTT) assay and supravital stain neutral red assay. Levels of IgG and IL 6 were measured by enzyme linked immunosorbent assay (ELISA). The proliferative response and IgG production of splenic lymphocytes in the presence of testosterone were lower than those of control cells. Testosterone may inhibit proliferative response and IgG production of splenic lymphocytes. The results also support that androgen may play an important role in immune regulations.
