心肌梗塞患者单核细胞-血小板聚合率的检测

摘要 目的 建立流式细胞术检测循环全血中单核细胞与血小板聚合率的方法。方法 全血与CD14-FITC和CD61-PE单克隆抗体共同孵育15min,加溶血素溶解红细胞,洗涤后用流式细胞仪检测。以CD14区分出单核细胞,检测单核细胞中CD61的表达率。CD14和CD61双阳性的细胞是单核细胞与血小板聚合物。结果 心肌梗塞组循环全血中单核细胞-血小板聚合率为25.8±8.3%,显著高于正常对照组6.2±3.5%。结论 流式细胞术可用于检测全血中单核细胞-血小板聚合率。单核细胞-血小板聚合率的检测对心肌梗塞患者具有一定的诊断价值。 Objective To establish a method for detection of circulating monocyte-platelet aggregates by flow cytometry. Method Whole blood was incubated with monoclonal antibodies CD14-FITC and CD61-PE for 15 min. Dissolved the RBC. Monocyte were first distinguished by CD14 and their expression detected by CD61. The double positive (CD14 and CD61) was considered as monocyte-platelet aggregates. Calculated the percentage of double positive cells among all CD14 positive monocyte. Results The monocyte-platelet aggregation rate was singnificantly higher in patient with miocardial infarction than that in the control group(mean±s%, 25.8±8.3% vs 6.2±3.5%). Conclusions Detection whole blood circulating monocyte-platelet aggregation rate by flow cytometry is conceivable. The percentage of monocyte-platelet aggregates in patients with miocardial infarction has certain diagnostic value.
