脱脂神经髓鞘诱导多发性硬化T淋巴细胞系增殖反应分析

摘要 目的 揭示中枢神经髓鞘脱脂过程在多发性硬化 (MS)发生发展中的作用。方法 以人脱脂神经髓鞘二次致敏诱导MS患者T淋巴细胞系 (MS TCL) ,用髓鞘碱性蛋白 (MBP)、蛋白脂蛋白 (PLP)及其合成多肽等抗原检测TCL的增殖反应。结果 MS组对 11种抗原均有较强反应 ,对 7种抗原的反应性与对照组间有差异 (P <0 0 5 ) ,两组总和平均阳性孔比较P <0 0 0 1(5 49± 5 31与3 10± 3 17比较 )。结论 PLP和MBP与MS发病有关 ;M87 10 6、P95 117、P40 6 0、P185 2 0 6等序列可能是致病抗原决定簇 ;MS病人体内可能有神经髓鞘异常脱脂。 Objective To identify the role of delipidated process in initial multiple sclerosis(MS).Methods T cell lines of multiple sclerosis(MS-TCL) were established using human delipidated myelin by short period culture in vitro and then their recognitions to MBP,PLP and their synthetic peptides,were examined.Results MS-TCLs displayed strong proliferations to 11 types of antigens.Moreover,there were statistically differences in recognitions to 7 types antigens between MS and control and so was between their general average positive wells( P< 0.001,3.41±4.83 vs 5.49±5.31).Conclusion Our results suggest that both PLP and MBP,and the myelin delipidated process might play important roles in MS pathological process.M87-106,P95-117,P40-60 and P185-206 might be the candidate epitopes to evocation MS and relapse.
