摘要
目的 探讨小剂量顺铂增加人Hela细胞对电离辐射敏感性涉及细胞周期动力学的机理。方法 将Hela细胞分为空白对照组、单纯放疗组、单纯化疗组和放疗加化疗组 ,用流式细胞仪测定细胞凋亡率和细胞周期变化 ,采用SP免疫组化法检测Hela细胞受作用后p5 3基因蛋白表达情况。结果 单纯放疗、单纯化疗均可诱导Hela细胞凋亡 ,而放疗加化疗组细胞凋亡率明显增加 ,经顺铂作用后 ,Hela细胞周期发生明显变化 ,表现为G1期减少、S期和G2 /M期增加 ,突变型 p5 3表达减弱。结论 小剂量顺铂可通过改变培养的人Hela细胞周期 ,诱导细胞G2 /M期延迟从而提高放疗敏感性 ,可能与其内在的 p5 3基因表达改变有关。
Objective To investigate low dose cisplatin in the enhance inout of the sensitivity of radiotherapy of human Hela cell lines throuth adjusting cell-cycle kinetics.Methods The Hela cells were divided into four groups(control group,radiation only,chemotherapy only,radiation and chemotherapy),flow-cytometry was introduced in the study of the cell cycles and apoptosis of Hela cells,and immunohistochemistry of p53 protein.Results Radiation and chemotherapy both could induce Hela cells apoptosis,cisplatin could obviously enhance Hela cells apoptosis induced by radiation,and cisplatin could change cell-cycle of Hela cell lines,G_1 phase cells decrease and G_2/M phase cells increase.Conclusion Low dose cisplatin can enhance the sensitivity of radiotherapy through change cell cycle of Hela cell lines.
