摘要
目的观察角蛋白CK10和CK19在组织工程皮肤和正常在体皮肤的表达,并探讨其差异表达的意义。方法用组织工程方法构建人全层皮肤,以正常在体皮肤为对照,免疫组化检测CK10和CK19在表皮层的表达。结果在组织工程皮肤中CK10表达阳性,CK19无明显阳性表达;正常在体皮肤CK10和CK19表达皆为阳性,其中CK19主要表达在基底膜附近。结论组织工程皮肤中表皮层细胞已经基本丧失了组织干细胞的特性,分化成熟。
Objective To examine the expressions of CK10an d CK19in tissue -engineered skin and normal human skin.Methods The bilayer skin replacement was constr ucted by tissue engineering method.The expressions of CK10and CK19were examined by immunocyto-chemistry.Results CK10was positively expressed in the bilayer skin replacement,while the expression of CK19was negative.Both CK10and CK19were positively expressed i n normal human skin,and CK19was main ly expressed around the basilar memb rane.Conclusion The epidermal cells in the bilayer skin r eplacement had differentiated and l ost the characteristic markers for k eratinocyte stem cells.
