摘要
目的 探讨纤溶酶原激活物抑制物 - 1 (PAI- 1 )启动子基因 4G/ 5G多态性与急性冠脉综合征(acutecoronarysyndrome,ACS)的关系。方法 应用聚合酶链反应 -限制性片段长度多态性 (PCR -RFLP)对 56例ACS病人及 76例正常人PAI- 1基因单核苷酸插入 /缺失状态进行分析 ,并研究PAI- 1基因多态性与血脂、体重指数 (BMI)、血压的关系 ,分析 4G/ 5G多态性与ACS的关系。结果 ①病例组与对照组比较 ,4G/ 4G基因型频率较高 (2 4 / 56和 1 5/ 76 ,P <0 0 1 ) ;病例组 4G/ 5G基因型频率明显低于对照组 (2 5/ 56和 48/ 76 ,P <0 0 5)。病例组 4G等位基因频率明显高于对照组 (0 652和 0 51 3) ,而 5G等位基因频率明显低于对照组 (0 348和 0 487,P <0 0 5)。②高密度脂蛋白胆固醇 (HDL)、低密度脂蛋白胆固醇 (LDL)两组差异无显著性意义 (P >0 0 5) ;病例组总胆固醇 (TC)、甘油三酯 (TG)、BMI、收缩压、舒张压均明显大于对照组 (P <0 0 1 ,0 0 0 1 ,0 0 1 ,0 0 5) ,差异有显著性意义。结论 PAI 1基因 4G/ 5G多态性与ACS密切相关 ,其 4G/ 4G基因型在ACS患者中所占比例较高 ,ACS患者显著高血脂、高BMI。
Objective To study the correlation between 4G/5G polymorphism located in the promoter region of plasminogen activator inhibitor 1(PAI 1)gene and acute coronary syndrome(ACS).Methods 4G/5G polymorphism were measured in 56 patients with ACS and 76 control subjects by a combination of polymerase chain reaction restriction fragment length polymorphism(PCR RELP)and direct AgNO 3 staining technique.Blood pressure?blood lipids and body mass index(BMI)were studied in the mean time.Results ①The frequencies of the genotype 4G/4G were higher in patients with ACS(24/56vs 15/76, P <0 01).The frequencies of 4G/5G genotype were lower in patients with ACS than control(25/56 vs 48/76, P <0 05).The frequencies of 4G allele were higher in patients with ACS(0 652 vs 0 513, P <0 05).②Those patients had higher total cholesterol(TC)( P <0 001),higher triglyceride(TG)( P <0 001),higher BMI and( P <0 01) higher blood pressure( P <0 05).Conclusion The frequencies of the genotype 4G/5G were higher in patients with ACS.4G/4G genotype is also correlated with ACS.The patients had higher lipids,higher BMI,higher blood pressure.[
