在以 PCR为基础的种子纯度鉴定和分子标记辅助选择育种中 ,DNA模板的制备常常成为主要的限速步骤 ,建立一种快速简便的微量 DNA模板制备方法有着十分重要的意义。本文介绍了一种以磨碎的水稻组织直接进行 PCR扩增的方法 ,该方法简单方便、稳定可靠 ,大大简化了 DNA模板制备过程 ,所制备的 DNA模板在 - 2 0℃保存 1个月后不影响
It is time consume to preparing DNA template for PCR based seed purity test and molecular marker assisted selection in plant breeding. The establishment of a simple method to facilitate DNA template preparation is very important. In this article, we reported a new method for preparation of preparation of DNA template for such prupose, which used drounded tissues of Oryza sativa as PCR template directly. This method is very simple and also stable. PCR templates prepared via this method could be stored at -20℃ for at least one month.
