摘要 目的 从体内体外两方面探讨BMP2 在牙髓损伤修复 ,尤其是牙髓细胞分化过程中的作用。方法 建立大鼠牙髓损伤修复模型和体外人牙髓细胞连续培养 ,采用免疫组织化学、MTT法和酶动力学等方法 ,检测BMP2 在体内牙髓组织损伤修复过程中的表达 ,并比较不同浓度rhBMP2 对体外人牙髓细胞连续培养各阶段细胞增殖活性及ALP活性的影响。结果 体内牙髓组织损伤修复过程中存在BMP2 的表达 ,牙髓组织损伤修复的不同阶段其表达部位和强度发生变化 ;rhBMP2 可提高体外培养人牙髓细胞的增殖活性 ,与其本身浓度有关 ,并可促进人牙髓细胞ALP活性 ,呈浓度依赖性 ;结论 BMP2 是参与牙髓损伤修复 ,尤其是牙髓细胞的增殖和分化的重要因子之一。 Objective To explore the roles of bone morphogenetic protein 2 (BMP 2) in the healing of injured dental pulp in vivo and in vitro . Methods The expression patterns of BMP 2 during the healing of injured dental pulp were studied with immunohistochemistry and the effects of recombinant human bone morphogenon 2 (rhBMP 2) on proliferation and differentiation of cultured human dental pulp cells were also studied in this experiment. Results The results clearly demonstrated that BMP 2 were expressed in the course of rat dental pulp healing, its expression sites and levels of their expression were changed with different phases; rhBMP 2 increased the proliferation of dental pulp cells and ALP activity in a dose dependent manner. Conclusion BMP 2 plays an important role in the healing of injured dental pulp, which may be due to promote the differentiation of dental pulp cells to odontoblasts.
