摘要
目的 探讨果糖二磷酸钠镁 (FDPM )抗大鼠心肌缺血损伤的作用是否与改善能量代谢有关。方法 大鼠左冠状动脉结扎 4h制备急性心肌梗死模型 ,于结扎后 5min分别自股静脉ivFDPM (4 5 ,90 ,15 0mg·kg- 1) ,硫酸镁 (10mg·kg- 1) ,1,6 二磷酸果糖钠 (130mg·kg- 1) ,硝酸甘油 (5mg·kg- 1)及等容量生理盐水 ,高效液相色谱法测定心肌缺血区能量物质含量 ,定磷法测定ATP酶活性。结果 FDPM(4 5 ,90 ,15 0mg·kg- 1)能增加缺血区心肌ATP ,ADP ,AMP及总腺苷酸含量和能荷 ;提高缺血区心肌Na+ ,K+ ATP酶、Ca2 + ATP酶和Mg2 + ATP酶活性 ;其具有 1,6 二磷酸果糖钠和硫酸镁的类似作用特点。结论FDPM通过增加缺血区心肌能量物质含量及提高ATP酶活性而产生心肌缺血保护作用。
AIM To explore whether the effect of sodium magnesium fructose diphosphate (FDPM) against ischemic myocardium injury in rats is related to improve energy metobolism. METHODS The acute myocardial infarction was induced by ligation of the left coronary artery for 4 h. FDPM(45,90,150 mg·kg -1 ), magnesium sulfate(10 mg·kg -1 ),fructose 1,6 diphosphate (130 mg·kg -1 ),nitroglycerine(5 mg·kg -1 )or normal saline was intravenously infused 5 min after the left coronary artery ligation. High energy phosphates (ATP, ADP, AMP) in the ischemic myocardium were assayed by high performance liquid chromatography and ATPase activity was assayed by colorimetric assay. RESULTS FDPM at the dose of 45, 90 and 150 mg·kg -1 iv increased the contents of ATP,ADP,AMP and total adenine nucleotide and energy charge, enhanced the activities of Na +, K + ATPase, Ca 2+ ATPase and Mg 2+ ATPase in ischemic zone of myocardium, suggesting that FDPM have similar characteristics of both fructose 1,6 diphosphate and magnesium sulfate. CONCLUSION FDPM exerts myocardial protection against ischemic injury by improving energy metobolism and ATPase activities.
