摘要 用机械破壁法直接提取来自垃圾填埋场滤液样本中的细菌总 DNA,以细菌通用引物对p A/p H和梭状芽孢杆菌属 ( Clostridium) Cluster 特异引物对 Clos71 5 /Clos1 45 2 ,针对细菌 1 6Sr DNA进行“网式”PCR扩增 ,获得 1 5 0 0 bp和 75 0 bp的扩增片段 ,选择一阳性扩增产物克隆和测序 .序列分析结果表明 ,垃圾填埋场滤液中含有降解纤维素细菌 ,且与 RDP数据库中的 Clostridi-um Cluster Celluloytic bacterial communities in landfill leachate were assessed with rRNA gene based technologies. Total bacterial DNA was extracted by the bead beating method, the bacterial universal primer pair pA/pH and specific primer pair Clos715/Clos1452 for Celluloytic Cluster Ⅲ were used to amplify 16S rRNA gene with “nested” PCR technology, and one of the positive “nested” PCR amplified products was coloned and sequenced. The sequence similarity analysis shows that there are celluloytic bacterial communities which are dramatic similar to Clostridium Cluster Ⅲ in landfill leachate.
