摘要
目的 :观察薯蓣皂苷元 (diosgenin ,Dio)的体内和体外抗肿瘤作用。方法 :体内试验采用 4种小鼠移植性肿瘤 (S 180 ,HepA ,U14和EAC)腋部皮下接种 ,于接种后次日开始给药 ,10日内每天给药 1次 ,计算各给药组瘤肿抑制率。体外试验采用 4种肿瘤细胞株 (MCF ,L92 9,A375 S2 ,HeLa) ,分别与不同浓度的药物共同培养 ,染色后计算其抑瘤率。结果 :①两种给药途径 (ig ,ip)下 ,Dio对S 180 ,HepA ,U14小鼠移植肿瘤具有抑制作用 ,抑制率在30 %~ 5 0 % ,而对EAC没有抑制作用 ;②体外Dio对L92 9,HeLa,MCF肿瘤细胞株具有抑瘤作用 ,IC50 分别为 1.2 ,18.2 ,19.8μg·mL-1,而对A375 S2细胞则效果不明显。结论 :Dio对S180 ,HepA ,U14等小鼠移植肿瘤和L92 9,HeLa ,MCF肿瘤细胞株具有明显抑制作用。
Objective: To investigate the antitumor activity of Diosgenin in vivo and in vitro. Method: S-180, HepA, U14 and EAC transplant mice were given Diosgenin ig or ip everyday for 10 days, from the next day when they were inoculated in axilla. Tumor growth inhibit rates were calculated .Four kinds of cells , MCF, L929, A375-S2 and HeLa ,were incubated respectively with Diosgenin in vitro. Tumor growth inhibit rates were also calculated . Result: In vivo, both ig and ip ,Diosgenin inhibited S-180,HepA,U 14 mice transplant tumor ,the inhibit rates being 30%~50%,but it did not inhibit the EAC mice transplant tumor.In vitro, Diosgenin inhibited L929,HeLa,MCF cell growth ,and IC 50 were 1.2, 18.2, 19.8 μg·mL -1 respectively, but it did not significantly affect A375-S2 cells. Conclusion: Diosgenin has an obvious antitumor activity on S-180, HepA, U14 transplant mice in vivo and L929,HeLa,MCF cells in vitro.
