摘要
目的 :研究卵巢上皮性癌细胞中细胞凋亡抑制蛋白的表达及作用。方法 :采用反义XIAP(X -linked细胞凋亡抑制蛋白 )腺病毒感染卵巢上皮性癌细胞系 ,用Hoechst染色计细胞凋亡率 ,用Western印迹法及扫描法检测XIAP的蛋白表达量。结果 :反义XIAP腺病毒抑制耐药性卵巢上皮性癌细胞的XIAP的表达 ,随感染时间的延长 ,细胞凋亡率升高。感染72h ,细胞凋亡率达 32 2 1% (P <0 0 0 1)。结论 :反义XIAP腺病毒抑制耐药性卵巢上皮性癌细胞XIAP的表达 ,诱导癌细胞凋亡 。
Objective:To study the roles of X-linked inhibitor of apoptosis protein(XIAP) expression in apoptosis of cisplatin-resistant ovarian epithelial cancer cell line (C13 *) Methods:C13 * cell line was transfected with adenovirus containing antisense XIAP gene artificially The apoptosis rate was counted by Hoechst 33248 staining method Following western blotting,XIAP expression was determined by Molecular Dynamic Phosphoimager Scanning Results:Antisense XIAP significantly inhibited XIAP expression in C13 * cell,at 72h infection point,XIAP expression was significantly decreased ( P <0 005),and apoptosis rate was significantly increased at 48,60 and 72h( P <0 001, P <0 0001, P <0 0001, respectively) Conclusion:Antisense XIAP can significantly inhibit the XIAP expression and induce apoptosis in cisplatin-resistant ovarian epithelial cancer cell line,indicating that XIAP may play a critical role in the pathway of apoptosis of drug-resistant ovarian epithelial cancer cell and that XIAP should be considered as a target gene for gene therapy
