缺血预处理对兔冠脉急性缺血再灌注期间血小板聚集率的影响

摘要 目的 研究缺血预处理 (IPC)对实验性冠脉急性缺血再灌注期间血小板聚集率的影响及腺苷的作用 .方法 建立兔在体心脏缺血预处理模型 (n=14 ) ,用智能血液凝仪测定血小板聚集率 ,高效液相色谱测定心肌组织及血液腺苷含量 .结果  IPC组缺血再灌注期间血小板聚集率无明显升高 ,与缺血前比较 (P>0 .0 5 ) ,与对照组比较差异显著 (P<0 .0 1) ,IPC组血液腺苷含量 (3.0 1± 0 .0 2 ) mg· L- 1 高于对照组(2 .2 5± 0 .19) mg· L- 1 (P<0 .0 5 ) ;心肌腺苷含量 (0 .85±0 .0 4 ) m g· g- 1与对照组 (0 .2 7± 0 .0 2 ) mg· g- 1比较差异显著 (P<0 .0 1) ,缺血前静滴腺苷可使血小板聚集率显著下降 .结论  IPC可使实验性冠脉急性缺血再灌注期间血液和心肌腺苷含量明显升高 ,血小板聚集率显著下降 ,而腺苷参与了IPC对心肌缺血的保护作用 . AIM To study the effect of ischemic preconditioning (IPC) on adenosine and platelet aggregation rate during coronary artery acute ischemic reperfusion in rabbits. METHODS IPC models were set up in rabbit hearts. Platelet aggregation rate was determined by using brain blood coagulator. Adenosine contents in myocardial tissue and blood were determined by high performance liquid chromatography. RESULTS Platelet aggregation rate did not increase obviously in the IPC group during ischemic reperfusion, and had no obvious difference as compared with that before ischemia ( P >0.05), but it was significantly different as compared with controll that of the group ( P <0.01), Blood adenosine content in the IPC group (3.01±0.02) mg·L -1 was higher than that in the control group (2.25±0.19) mg·L -1 ( P <0.05). Myocardial adenosine contents in the IPC group (0.85±0.04) mg·g -1 was obviously differet as compared with that of the control group [(0.27±0.02) mg·g -1 ( P <0. 01)]. Intravenous drip of adenosine before ischemia could obviously reduce platelet aggregation rate. CONCLUSION IPC could significantly increase blood and myocardial adenosine contents, and obviously reduce platelet aggregation rate during coronary artery acute ischemic reperfusion in rabbits. The results suggest that adenosine participates in the protective effects of IPC on myocardial ischemia.
