家兔胸部火器伤后肺组织中IL-1β表达变化(英文)

摘要 目的 用免疫组织化学方法检测兔胸部火器伤后肺组织不同部位 IL - 1β的表达 ,探讨其伤后表达变化规律及在局部组织损伤中的作用 .方法  6 4只家兔随机均分 8组 ,除正常对照组外均用 5 .5 6 mm小口径步枪距离 6 m射击兔右下胸第八肋间锁骨中线外 1cm处造成前后胸贯通伤 ,经指袢排气及消毒缝闭胸壁伤口等简单救治 .7组致伤家兔存活者分别于伤后 0 .5 ,1,6 ,2 4 h,3,7d,1mo活杀 ,分别于伤肺距伤道 2 mm处、1cm处、临叶、与伤道对应的对侧肺处取肺组织标本 ,行 HE染色及免疫组织化学方法检测肺组织病理损伤及 IL- 1β的表达 .结果 伤后 0 .5 h肺组织中 IL- 1β表达即升高 (P<0 .0 1) ,6 h已达高峰 ,持续数日 ,1w后降低 ,1mo后与伤前无明显差别 .不同部位肺组织中 IL - 1β表达部位、变化趋势基本相同 ,而表达强度有所不同 ,原发伤道 >挫伤区 >临叶 >对侧叶 .结论 胸部火器伤后家兔肺组织中 IL - 1β表达迅速升高 ,离原发伤道距离越近 ,IL- 1β表达升高越显著 .肺组织 IL- 1β表达升高水平与该处肺组织出血、水肿程度成正相关 (r=0 .9993,P<0 .0 5 ) ,IL - AIM To investigate and discuss the regularity of the expression of interleukin 1β in rabbit's lungs after being shot through the thoracic cavity. METHODS Sixty four rabbits were divided into 8 groups randomly, shot at the point 1cm away the middle line of clavicula in the 8th intercostals spatium with 5.56 mm caliber rifle gun, 7 of 8 groups rabbits were wounded and 1 group was normal group, 2 mm 3 lung tissues were cut from such spots like 2 mm and 1 cm far from the prinary wound tract (PWT), adjoining lobe and opposing lobe respectively at the time 0.5, 1, 6, 24 h, 3, 7, 30 d after being shot, and the pathological changes of lung tissue and expression of IL 1β by HE staining and immunohistochemistry respectively. RESULTS Expression of interleukin 1β improved from the time of 0.5 h after being shot , reached peak at 6 h and kept on for days and then reduced from 1 wk after being shot, recovered to normal 30 d after being shot. Different expression appeared in different areas, such as PWT of wounded lobe > contusion area of wounded lobe > adjoining lobe > opposing lobe ( P <0.01). There was a good consistent between HE and immunohistochemistry in showing the degree of wound. CONCLUSION The nearer to the PWT, the higher the expression of interleukin 1β in lung tissue, and the expression of interleukin 1β in lung tissue has a good relationship to the degree of hemorrhage and edema in lung tissue and interleukin 1β plays an important role in the inflammation in rabbits thoracic firearm wound.
