摘要
目的 探讨人卵巢癌细胞体外刺激、诱导的肿瘤特异细胞毒性T淋巴细胞 (CTL)对靶细胞的杀伤活性。方法 应用建株的人卵巢癌细胞 (OVA 319) ,在体外定期刺激、诱导肿瘤患者外周血淋巴细胞 ,经微量淋巴细胞毒试验和流式细胞术分别测定扩增的淋巴细胞组织相容性白细胞抗原 (HLA)I类分子表型和膜表面免疫标志物 ,并通过 4 甲基偶氮唑盐 (MTT)法分析其对肿瘤细胞的杀伤活性。结果 经卵巢癌细胞刺激、诱导 3周后扩增的淋巴细胞主要为T细胞抗原受体 (TCR) α、β及CD8阳性的CTL ,具有明显特异性杀伤肿瘤细胞的作用 ,并发现经OVA 319细胞刺激、诱导后不同个体的卵巢癌患者CTL的杀伤活性与效靶细胞的HLA A、B位点相同数成正比。结论 肿瘤细胞体外激活的CTL具有特异杀伤靶细胞作用 ,提示CTL特异杀伤作用与刺激细胞、靶细胞及本身CTL表型的HLA A。
Objective To appraise the tumor specific cytolitic T lymphocytes(CTLs), which were stimulated and induced by human ovarian cancer cells in vitro and its activity of killing the target cells. Methods The peripheral blood lymphocytes from cancer patients were periodically stimulated and induced in vitro using human ovarian cancer cell clone (OVA 319). Phenotypes of histocompatibility leukocyle antigeno(HLA) class I molecules and immune markers of the surface membrane of expanded lymphocytes from these patients were determined by micro quantitative cytotoxic test and FACS technique respectively, and the tumor specific CTL activity of killing target cells was analyzed by MTT method.Results The lymphocytes which were stimulated and induced by human ovarian tumor cells for three weeks manifested mainly T cell receptor(TCR)αβ、CD8 positive T cells (CTL) with specific cytotoxic effect on the tumor cells. It was shown that the killing activity was positively correlated with the same number of HLA A, B loci.Conclusion These findings suggest the close relationship of CTL specific killing effect to the HLA A, B loci of stimulated cells , target cells and CTL themselves.
