CYP2C19基因多态性与云南汉族人群冠心病的相关性研究 被引量:10

摘要 目的 研究细胞色素P450 (CYP)2C19基因多态性与云南汉族人群冠心病的相关性。方法 随机选取2012年9月—2013年12月在昆明医科大学第一附属医院心内科住院且无血缘关系的云南汉族人群127例作为研究对象,采用基因芯片法检测所选血液样本DNA 中CYP2C19基因序列单核苷酸多态性位点CYP2C19*1、CYP2C19*2、CYP2C19*3的基因型,分析63例冠心病组和64例对照组的基因多态性,从而分析基因多态性与冠心病的相关性。结果Logistic回归分析结果:除了年龄、性别、高血压、糖尿病、高胆固醇血症等传统危险因素之后,CYP2C19*1/*2基因突变者患病风险明显增加[OR =5.24,95% CI (2.00~13.72)],CYP2C19*1/*3基因突变者患病风险亦明显增加[OR=4.54,95% CI (0.55~37.08)]。结论CYP2C19基因多态性是云南汉族人群冠心病发生的重要危险因素。CYP2C19基因多态性可以增加云南汉族人群患冠心病的发生风险。 Objective To investigate the correlation between gene polymorphism of cytochrome P450 (CYP)2C19 and coronary heart disease (CHD)in Yunnan Han population.Methods One hundred and twenty seven Yunnan Han population were randomly selected from September 2012 to December 2013.Using gene chip assay,the genotypes were detected in the selected blood sample with genotype CYP2C19 gene sequence of single nucleotide polymorphisms CYP2C19 * 1,CYP2C19 * 2,CYP2C19 * 3 genotype.The gene polymorphisms were analyzed in CHD group (n = 63)and control group(n = 64).The correlation of gene polymorphism and CHD were analyzed.Results Logistic regression analysis showed that in addition to age,sex,hypertension,diabetes,hypercholesterolemia and other traditional risk factors,CYP2C19* 1/* 2 gene mutation significantly increased risk of CHD[OR =5.24,95% CI (2.00 to 13.72)],CYP2C19* 1/ * 3 gene mutation was also significantly increased the risk [OR = 4.54,95% CI (0.55 to 37.08)].CYP2C19 gene polymorphism was an important risk factor for coronary heart disease in Yunnan Han population.Conclusion CYP2C19 gene polymorphism may increase the risk of coronary heart disease in Yunnan Han population.
