“活血化瘀、行气止痛”大法对输卵管炎性不孕大鼠模型中TLR4受体信号转导通路作用机理的研究 被引量:5

摘要 目的:探讨“活血化瘀、行气止痛”大法对输卵管炎性不孕大鼠模型中TLR4受体信号转导通路的作用机理,进一步说明输卵管炎性不孕与TGF-β(转化生长因子)、γ-IFN(干扰素)、NF-κB(核转录因子)之间的关系,以及中药膈下逐瘀冲剂对其表达的影响。方法:选取鼠龄为8~12周成熟大鼠120只,混合菌造模后,将大鼠随机分为正常组、模型组、西药组、膈下逐瘀冲剂高、中、低剂量组,每组各20只,适应性喂养7d后,进行灌胃猝死取血,免疫组化法观察TGF-β、γ-IFN、NF-κB蛋白的表达。结果:与模型组相比,中药膈下逐瘀冲剂中剂量组能够明显抑制TGF-β、γ-IFN、NF-κB蛋白的表达,达到消除炎症的目的。结论:膈下逐瘀冲剂是治疗输卵管炎性不孕的有效方剂,为输卵管炎性不孕发病机理提供了理论依据。 Objective: To study function mechanism of the Huoxue Huayu Xingqi Zhitong therapy on TLR4 receptor signaltransduction pathway in fallopian tubal inflammatory infertility rat model, and further illustrate the relationship among salpingitis infertilityand transforming growth factor, interferon, nuclear transcription factor etc., and the infl uence of the Gexia Zhuyu granules onthe expression. Methods: 120 8~12-week-old rats were selected; after mixed bacteria modeling, all rats were randomly divided intothe normal group, the model group, the western medicine group, and Gexia Zhuyu granules of high, medium and low dose groups, 20cases in each. All rats received adaptive feeding for 7 days, then received blood drawn after sudden death because of gavage. TGF-β,γ-IFN and NF-κB protein expression were observed by immunohistochemical staining. Results: Compared with the model group,TGF-β, γ-IFN and NF-κB protein expression in the Gexia Zhuyu granules middle dose group were significantly inhibited, in order toeliminate inflammation. Conclusion: The Gexia Zhuyu granules were effective on salpingitis infertility, and can provide a theoreticalbasis for the pathogenesis of salpingitis infertility.
