TLR-9在胰腺癌细胞化疗中对吉西他滨耐药性的影响

Effect of TLR-9 on drug resistance of pancreatic cancer cells to gemcitabine

目的:探讨TLR9影响胰腺癌PANC-1细胞对吉西他滨的耐药性。方法:将PNCA-1细胞随机分为3组。A组给予生理盐水+吉西他滨,B组给予TLR9激动剂+吉西他滨,C组给予TLR9抑制剂+吉西他滨。检测3组细胞凋亡水平及bcl-2蛋白的表达。结果:B组凋亡率小于A组与C组,差异具有统计学意义(P<0.05),而C组凋亡率大于A组,差异具有统计学意义(P<0.05)。A组Bcl-2的表达与B组比较,差异具有统计学意义(P<0.01)。B组Bcl-2的表达与C组比较,差异具有统计学意义(P<0.01)。A组Bcl-2的表达与C组比较,差异具有统计学意义(P<0.05)。结论:激活TLR9可明显增加胰腺癌PNAC-1细胞株对吉西他滨化疗的耐药性,抑制TLR9可增加胰腺癌细胞对吉西他滨的敏感性。

Objective:To investigate the effect of TLR9 on the drug resistance of pancreatic cancer cells PANC-1 cells to gemcitabine.Methods:PNCA-1 cells were divided into three groups randomly.A group was given normal saline + gemcitabine;B group was given TLR9 agonist + gemcitabine group;C group was given TLR9 inhibitor + gemcitabine.The level of cells apoptosis and the expression of Bcl-2 protein in the three groups were detected.Results:The apoptosis rate of B group was less than that of C group and A group,and the difference was statistically significant (P<0.05).The apoptosis rate of C group was higher than that of A group,and the difference was statistically significant (P<0.05).There was no statistically significant difference of expression of Bcl-2 between group A and group B (P<0.01).There was no statistically significant difference of expression of Bcl-2 between group B and group C (P<0.01).There was no statistically significant difference of expression of Bcl-2 between group A and group C (P<0.01).Conclusion:The activation of TLR9 could increase the drug resistance of pancreatic cancer PNAC-1 cell line to gemcitabine chemotherapy significantly.The inhibition of TLR9 could increase the sensitivity of pancreatic cancer cells to gemcitabine.
