摘要
目的:探讨前列腺术后前列腺部尿道创面修复的细胞来源。方法:12只成年雄性中华田园犬,随机分为两组。实验组接受经膀胱前列腺切除术,对照组仅膀胱切开后缝合。于术后3 d、1周和2周处死动物留取前列腺部尿道标本,采用HE染色观察前列腺部尿道病理学改变,免疫组化检测CK34、PSA表达。结果:HE染色示术后1周散在新生上皮形成,术后2周时基本覆盖创面。免疫组化显示术后3 d前列腺部尿道创面未见CK34、PSA表达;术后1周创面出现新生散在、岛状分布的CK34阳性细胞,未见PSA阳性细胞;术后2周新生上皮覆盖创面,CK34表达阳性,与正常尿道上皮相延续,但仍未见新生PSA阳性细胞。结论:创面下残余前列腺组织是前列腺切除术后修复来源,基底层CK34阳性细胞是参与该修复的细胞来源,而PSA细胞未直接参与。
Objective:To explore the cells involving in re-epithelialization of prostatic urethra after prostatectomy. Methods:Twelve male canines were randomly divided into two groups and underwent transvesical prostatectomy or sham operation respectively.Wounds were observed with HE and immunohistochemical staining at day 3 , week 1 and 2. Results : At day 3 , CK34 and PSA expressionwere negative. CK34 was detected in newly generated epithelium at week 1, but PSA was not. At week 2 , germinated epitheliumformed by CK34 positive cells covered the wound, extending to urothelium. However, there was no PSA positive cell in newly generatedepithelium. Conclusion: CK34 positive cells involved in re-epithelialization of prostatic urethra after prostatectomy. PSA positivecells may not play a direct role in that process.
