响应面法优化提取荚果蕨总黄酮的工艺研究

摘要 采用响应面法优化荚果蕨总黄酮的热浸提取工艺。在单因素试验的基础上,采用响应面法考察了提取温度、提取时间、液料比和乙醇浓度对总黄酮提取率的影响,并利用DesignExpert 8.0.6软件和Box-Behnken中心组试验设计原理对各工艺参数进行了回归分析。结果表明:提取温度是对响应值影响最大的因素,最佳提取条件为提取温度73℃、提取时间158min、液料比27、乙醇浓度74%,在此条件下,荚果蕨总黄酮的提取率为1.06%,与理论值相比略有增加,相对误差为0.95%,验证了回归模型的有效性。响应面法对荚果蕨总黄酮的提取条件优化合理可行,为荚果蕨总黄酮的开发与利用提供了理论依据。 Response surface methodology w,as employed to determine optimum conditions for extraction of total flavonoids fromMatteuccia struthiopteris ( L .) Todaro. The temperature on extraction, extraction time, liquid-solid ratio and the concentration ofethanol were chosen to conduct the single factor experiments. And regression analysis on various process parameters was carried outaccording to the Design-Expert 8.0.6 software and the Box-Behnken center combination experimental principles. The results showedthat the largest effect on response was the extraction temperature, and the optimum extraction condition was : extraction temperature73丈, extraction time 158min, liquid-solid ratio 27 and the concentration of ethanol 74% , respectively. Under these conditions, theactual value was 1 .0 6 % , which had a slight increase as compared to the predicted value, and the relative error was 0 .9 5 % ,indicating that the regression model fitted well with the experimental data. This study could provide a theoretical direction forextracting flavonoids from Matteuccia struthiopteris ( L .) Todaro on a large scale.
