复方苦参注射液对人肺腺癌A549细胞株增殖、乙酰肝素酶表达及侵袭能力的影响

Effects of compound radix sophora flavescentis injection on proliferation、migration and the expression of Hpa in human lung adenocarcinoma cell line A549

目的：通过体外研究探讨复方苦参注射液对人肺腺癌A549细胞株增殖、乙酿肝素酶(heparanase，Hpa)表达的影响及机制。方法:WST-1法检测不同浓度复方苦参注射液对A549的增殖抑制作用；S-P法测定各组干预48h后细胞内Hpa的表达;Matrigel体外侵袭试验检测各组细胞侵袭能力。结果:①0.1~0.5mg/ml浓度的复方苦参注射液作用人肺腺癌A549细胞达48h开始对细胞增殖有抑制作用，且呈浓度、时间依赖性;相同浓度时干预48h对细胞的增殖抑制作用优于72h。②复方苦参注射液作用肺腺癌A549细胞48h后，药物在0.1~0.5mg/ml浓度区间内对A549细胞核内Hpa表达有抑制作用。③复方苦参注射液作用肺腺癌A549细胞48h后，0.2~0.5mg/ml浓度区间内对A549细胞侵袭基底膜均有抑制作用。结论:本研究结果显示复方苦参注射液能通过抑制肺腺癌A549细胞表达Hpa、抑制A549细胞侵袭基底膜而发挥抗肺腺癌作用。

Objective To investigate the possible effects and mechanisms about compound radix sophora flavescentis injectionon proliferation and the expression of heparanase( Hpa) of human lung adenocarcinoma cell line A549 in vitro. Method Using theWST - 1 to detect the effect on cell growth inhibition rates of all groups after administration of compound radix sophora flavescentisinjection;The expressions of Hpa were dectected by immunocytochemical SP assay in each group after 48h period;The invasion abilityof cells is detected by Matrigel after 48 h period. Results (l)The proliferation of A549 cells could not be inhibited by Compoundradix sophora flavescentis injection for 24 h,however,it could be inhibited by compound radix sophora flavescentis injectionfor 48 and 72 h. And the energy of inhibition had relationship on concentration of compound radix sophora flavescentis injectionwith 48 h. Compared with 72 h group, the effect on proliferation inhibition was better in 48 h group.② Compound radix sophoraflavescentis injection could restrain the expression of Hpa of lung adenocarcinoma cell line A549 with the concentration of 0. 1 ~0. 5 mg/ml for 48 h . ③ Compound radix sophora flavescentis injection could restrain the migration of lung adenocarcinoma cellline A549 with the concentration in 0. 2 ?0. 5 mg/ml for 48 h. Conclusion This research shows Compound radix sophora flavescentisinjection has the capacity to inhibits the growth of A549 cell and the effect has the relationship on restraining the expressionof Hpa and the migration.
