摘要
目的:建立兔耳增生性瘢痕模型,通过局部注射人参皂苷Rb1,观察增殖细胞核抗原(Proliferating Cell Nuclear Antigen PCNA)的表达变化。方法:取8只新西兰白兔,每只兔耳6处创面,建立增生性瘢痕模型,设空白组,地塞米松组,生理盐水组,人参皂苷Rb1组。待伤口上皮化后,局部注射药物,每3d 1次,共三次,空白组不予处理。给药后第1、2、4、8周切除标本行MASSON染色观察瘢痕情况及瘢痕增生高度,免疫组化法检测PCNA表达。结果:注射后4、8周,与生理盐水组比较,地塞米松及人参皂苷Rb1组瘢痕均改善明显,瘢痕增生高度降低(P<0.05),PCNA表达降低(P<0.05)。结论:人参皂苷Rb1可有效抑制细胞增殖,改善兔耳增生性瘢痕增生。
Objective To observe the effect of ginsenoside Rb1 on hypertrophic scars of rabbit`s ear and test the expression of Proliferating Cell Nuclear Antigen(PCNA). Methods 8 rabbits were chosen randomly and 6 wounds were made in each rabbit ear to establish hypertrophic scars(HS).After the wounds epithelialization,the HS were separated into 4 groups: Blank group,Dexamethasone group,saline group,Ginsenoside Rb1 group. The administration was done once every 3 days for 3 times.On 1st,2nd,4th,8th weeks after the treatment,2 rabbits`s samples were resected randomly to test Masson staining and detect PCNA expression by immunohistochemistry. Results On 4th and 8th weeks after administration,compared with saline group,the scars of dexamethasone and ginsenoside Rb1 group were improved significantly,Scar elevation index reduced(P <0.05),the expression of PCNA decreased(P <0.05). Conclusion Ginsenoside Rb1 could effectively inhibit cell proliferation and improve scar hyperplasia.
